Primer on physiological techniques Different recording techniques:-Intracellular recording-Extracellular recording-Imaging Intracellular recording Record inside the cell using glass (why not metal?) electrodes. $$ This technique allows the experimenter to record subthreshold responses, and to understand mechanisms behind a cell's response properties. Whole-cell recording is a modern form of intracellular recording. This low-noise technique was the first to prove the existence of ion channels in cells. $$ This technique allows the experimenter to introduce chemicals into the cell and manipulate the cell's milieu. XX Cannot record from an awake animal using this technique (so far). XX Low yield, back-breaking work Extracellular recording As the name suggests, this technique entails the use of a metal (why not glass?) electrode that stays close to but outside the cell. Using this technique, one may record from: anesthetized, paralyzed animals, and from $$ awake monkeys $$ Stable recording $$ Excellent (almost as good as intracellular) single-unit isolation XX The techniques described above have one thing in common: only one neuron (or a few, at most) can be recorded simultaneously. Other techniques have been developed to record from a population of neurons.
